The shearing process is mainly in the inactivated vaccine sector, with vaccines of Water-In-Oil structure, Oil-In-Water structure and biphasic system as the main products.
Among them, Water-In-Oil as the most commonly used dosage form is most common in poultry and peptide oil seedlings.
Work Flow1. Oil phase preparation.
2. Water phase preparation.
3. The oil and water are emulsified at room temperature to form a Water-In-Oil, Oil-In-Water or two-phase system.
4. After emulsification, it is directly transferred to the canning area for canning.
Problem1. The particle size distribution of oil phase and water phase after emulsification cannot be controlled uniformly, and the particle size is different.
2. Wide particle size distribution, unstable emulsion droplets, short storage time.
3. Part of the water phase material sinks, causing the upper material to be too transparent and clear, and the system is not uniform.
4. The phenomenon of batch demulsification occurs.
Disadvantages of Traditional Emulsification:
1. Low-speed stirring inside the tank and single-stage large-flow emulsification outside the tank result in insufficient emulsification efficiency. Generally, 6-8 cycles can complete the emulsification process.
2. With the in-tank shearing process, the maximum batch size cannot exceed 2T, and the production batch increase cannot be achieved.
SolutionIn the field of vaccines, FLUKO adopts in-tank pre-emulsification process and cooperates with inline shearing circulation process for product preparation.
1. The emulsification tank is equipped with a high-speed pre-emulsification equipment to pre-mix and pre-emulsify the materials in the tank as a whole; with the high-speed inline shearing equipment, the materials are subjected to high energy density in the narrow working cavity to achieve maximum probabilistic emulsification process and to reach narrow distribution and small particle size.
2. The pre-emulsion equipment in the tank is a structure that does not require external coolant, and the equipment in the tank is isolated from the external environment through a special seal.
3. The new high tip speed inline shearing equipment achieves a maximum tip speed of 40M/s. The three sets of special shearing work heads in the cavity make the particle size distribution more uniform and close to nanometer size.
4. By setting the inline mixing unit, the powder can be added inline, and the powder can be introduced into the core mixing area immediately, which greatly improves the mixing efficiency.
